Any help or advice would be great...especially from people who have done this before. Duration? Cost?

It's a bold, long and adventurous trip but since Afghanistan is on your route, I'd advise now is not the best time to risk going through it. Even during peacetime(?), Afghan transportation infrastructure is one of the most dangerous in the whole wide world. Now you have Taliban, NATO bombing, mines and IEDs to deal with.

Yes that Iran-Afghanistan-Pakistan part would be something you would want to avoid. You can go from Iran to Quetta and avoid Afghanistan that way. However Iran is now under sanctions while Pakistan is not very safe either.
