It's not like the existing one is working double tides.
Antonov itself can't afford to finish the second one, and nobody else is willing to invest. Although there's been some limited interest from Volga-Dnepr and Russian ministry of defence, and maybe from somebody else. Meanwhile...

That's an old frame, if they ought to compete they need a complete redesigned monster, more efficient engines alone are not enough to reduce expenses, when avionics are outdated.

Still, the main reason is market demands. Antonov has been asked many time about second Mriya. Main answer is weak demand. And as you pointed above, new Mriya needs to be redesigned to fit up to date fuel efficiency ans reduce cost operation. It's quite a big R&D.

I am wondering if the A380 would provide a viable base to be modified into an alternative to the AN225. Any insights especially in terms of technical feasibilities?

Given small demand for "monster cargo planes" and very huge R&D cost, it won't happen soon.

With modern (read western) engines, they could probably lift the same weight with two as opposed to the six they have currently.

Just 2 engines? It won't handle it. Minimum 4 engines.

Its six engines produce about 310,000 lbf thrust, a pair of GE90-115Bs (the most powerful fan on earth) would max out at about 230,000 lbf of thrust, so it doesn't seem plausable. 4 would be very doable, but nobody is even thinking about a fan that does 150,000 lbf thust. A redesign for 4 GP7200s, GEnXs or Trent 900s would be able to comfortably power a an-225.
