There are two Frankfurts in Germany: Frankfurt an Main and Frankfurt an der Oder. Frankfurt is the name of the city, but the rest refers to be river Main / Oder.
'Am Main' means that Frankfurt is located by the river Main.

Frankfurt is located on the river Main which is flowing into the bigger river Rhein. "Main" is pronounced like English "mine" and NOT like "main". Though English uses Latin characters it doesn't use Latin pronouncation like German does and that leads to alot of confusion when English speakers try to read German.
